Mucoadhesive polymer formulations represent an advanced approach in biomedical engineering aimed at improving therapeutic outcomes through prolonged retention at mucosal surfaces. By adhering to mucus layers, these systems enhance localized drug delivery, increase bioavailability, and reduce dosing frequency. Their ability to maintain intimate contact with biological tissues makes them especially valuable for mucosal routes such as oral, nasal, ocular, and vaginal administration [1]. The engineering of mucoadhesive systems relies on understanding the mechanisms of mucoadhesion, including hydrogen bonding, electrostatic interactions, wetting, and polymer chain interpenetration. Polymer characteristics such as molecular weight, flexibility, charge density, and functional groups critically influence adhesive strength. Both natural polymers (e.g., chitosan, alginate) and synthetic polymers (e.g., carbomers, polyacrylic acid) are widely investigated for their tunable properties [2]. Mucoadhesive polymers are formulated into diverse dosage forms, including hydrogels, films, tablets, microspheres, and nanoparticles. These formulations enable controlled and sustained drug release while protecting active agents from enzymatic degradation. Advances in nanotechnology have further expanded their applications, particularly in targeted drug delivery and improved patient compliance [3]. Despite their promise, challenges remain regarding mucosal variability, long-term stability, toxicity, and large-scale manufacturing. Ongoing research focuses on smart and stimuli-responsive mucoadhesive systems to overcome these limitations. Overall, engineered mucoadhesive polymer formulations hold significant potential for future biomedical and pharmaceutical applications [4].
